黑曲霉原生质体的制备及再生 PREPARATION AND REGENERATION OF THE PROTOPLAST FROM ASPERGILLU NIGER 曹军卫 Cao Junwei first-author 研究了黑曲霉菌丝原生质体形成和再生的条件,讨论了菌龄,酶的浓度和酶的种类对原生质体释放的影响,以及不同菌龄菌丝原生质体再生能力的差异。发现处于对数生长期和稳定期的菌丝均能被酶解,得到较大量的原生质体。采用蛋清溶菌酶和蜗牛酶酶解黑曲霉,二者效果相近。在我们的实验范围内,蛋清溶菌酶的使用浓度则是越高越好。当用不同菌龄菌丝的原生质体进行再生时,发现菌龄较长的菌丝制得的原生质体,较容易再生。同时,在显微镜下对原生质体和再生菌丝的形态进行了观察。 The conditions of the formation and regeneration of mycelial protoplasts from Aspergillus niger were tested. The effects of mycelial ages, concentrations and kinds of different enzymes on release of the protoplasts were discussed, as well ar the differences of the regeneration ability of the mycelial protoplasts at different ages. It was found that the mycelia at exponential and stationary growth phases could be digested and a large amount of protoplasts released. Similar results were obtained when A. niger was lysised with the egg-white lysozyme and the snail enzyme. In our experiment the higer coneentration of the egg-white lysozyme used, the better the result it has. It seems to us that proloplasls prepared from older-aged mycelia regenerated easily. Mophology of the protoplasts and the regenerated mycelia were also observed with the mieroseope. 1984-04-01 2021-04-01 4
