亚硒酸钠诱导HeLa细胞凋亡机理 Sodium Selenite-Induced HeLa Cells Apoptosis 梁菊 1 first-author 邱芬 1 朱建发 1 刘志洪 1 蔡汝秀 1 沈萍 2 武汉大学化学与分子科学学院 武汉大学化学与分子科学学院 武汉大学生命科学学院 武汉大学生命科学学院 对亚硒酸钠诱导人宫颈癌细胞(HeLa)凋亡进行了研究,分别用荧光倒置显微镜和流式细胞仪进行凋亡形态分析和DNA片断分析.研究发现Na2SeO3对HeLa细胞的凋亡作用呈现浓度依赖性.当Na2SeO3的浓度分别为21,42,63μmol/L时,流式细胞仪检测到的细胞凋亡率分别是18.9%,33.0%,58.9%.为了进一步说明凋亡过程的分子机理,分别使用Frua-2/AM和DCFH-DA两种荧光染料检测了用Na2SeO3诱导的细胞内游离Ca2+含量和活性氧(ROS)水平的变化,发现在凋亡过程中伴随有明显的胞内Ca2+和ROS水平升高,且呈现Na2SeO3浓度相关性.这表明Ca2+和ROS在Na2SeO3诱导的HeLa细胞凋亡过程中起着重要的信号传导作用.
