Myristylated membrane protein(MMP) gene was cloned and identified from Rana grylio virus(RGV).It is 972 bp long and encodes a protein of 323 aa with a predicted molecular mass of 35×103.Alignment of the deduced amino acid sequence of RGV MMP showed that they had the homology degree of more than 64% with other Ranavirus. The recombinant prokaryotic expression plasmid containing this fragment was constructed and expressed an expected 53×103 fusion protein. The antiserum of MMP protein was produced. Its temporal expression pattern during RGV infection course was characterized by RT-PCR and Western blot analysis. It begins to transcribe as early as 4 h postinfection(p.i.) and translate after 6 h p.i.
which remains detectable at 48 h p.i.Using drug inhibition analysis by a de novo protein synthesis inhibitor(cycloheximide) and a viral DNA replication inhibitor(cytosine arabinofuranoside)
RGV mmp was classified as a late(L) viral gene during the in vitro infection.
