Based on the successful culturing of Dunaliella salina in the laboratory
high quality cDNA library from salt-tolerant Dunaliella salina cultured at 1.5 mol/L NaCl was constructed by using Takara cDNA Library Construction Kit.According to the initial characterization
the titer of primary cDNA library was 1.2 106 cfu/mL
and the rate of recombinant was above 95%.The colony PCR results showed that the lengths of most inserts were larger than 500 bp.Initial Expressed Sequence Tag(EST) analysis indicated that there were large amount of novel genes in the Dunaliella Salina genome.Random sequencing of 60 ESTs and subsequent bioinformatics analyses revealed that about one-third of the ESTs represented the novel genes which were never characterized before.The discovery of these novel ESTs(genes) laid the foundation for further screening salt-tolerant genes from Dunaliella Salina.
